Optimization of Culture Conditions for Cellulase Production by Wild Strain of Aspergillus Niger Using Sugar Cane Bagasse as Substrate

Abstract


Studies were conducted to produce cellulase by cellulolytic fungi using sugar cane bagasse as substrate. Aspergillus niger was isolated from domestic wastes and assessed for cellulase production using sugar cane bagasse as substrate. The effect of factors such as incubation period, temperature, initial pH and inoculums size were analysed and optimized. Cellulase production by the A. niger was found to be optimum/ highest with an incubation temperature of 40oC, pH 6.0, inoculums size of 1.0 × 106 spores/ml and incubation period of 5 days. The cellulases produced were partially purified using ammonium sulphate. Higher cellulose yield was found with the partially purified alkali treated sugarcane bagasse enzyme solution (0.74225U/mL) compared to the alkali treated sugarcane bagasse crude enzyme solution (0.3599U/mL).
